Cornwall made 189 for 8 with Greg Smith (Penzance CC) top scoring on 35 for the home county.
South African Smith, 36, is director of cricket development and head coach at Penzance CC.
Buckinghamshire were bowled out for 154 in the 35th over.
Cornwall now face a quarter-final trip to Berkshire on Sunday June 30.
Berkshire - who, like Cornwall, had a first round bye - beat Dorset
• The Cornwall Choughs Development XI beat Somerset Academy at North Perrott CC by seven wickets.
Cornwall Over 50s lost their ECB game, also against Somerset, at Taunton Deane CC by five wickets.
