A career-best 158 from opener Jack Mynott helped Wiltshire to establish a useful first innings lead of 69 before Berkshire slumped to 118 for nine in their second innings, just 49 ahead.
Seamer Sam Freestone and off-spinner Joe King shared eight wickets between them and sent Berkshire tumbling from 73 for two to 98 for eight. Freestone has taken nine wickets in the match so far and 16 in his first two games in the competition.
Toby Greatwood, who began the Championship season with a maiden century against Oxfordshire, held on to the close with last man Lewis Sharp but a first defeat in the competition since Cheshire beat them at Nantwich in 2022 now looms for Berkshire.
Wiltshire’s last victory over Berkshire came in 2014 at Falkland when King took eight wickets in the match, six in the first innings.
A win at Henley would not only improve Wiltshire’s chances of avoiding an immediate return to Division Two but also set up a potential Western Division title-decider with Devon at the South Wilts club in Salisbury starting on Sunday.
They owe their strong position in this match to Mynott who batted for four-and-a-half hours in the fourth Championship century of his career with excellent support from Warwickshire’s Jake Lintott (71) and Jake Goodwin (51).
Sussex batter Harrison Ward transformed Oxfordshire’s fortunes with an unbeaten century against Herefordshire at Thame.
Oxfordshire trailed by 171 on first innings but ended the day 239 in front with three wickets intact thanks to Ward’s 104 not out.
It was a second century in the competition for Ward, who is on a T20-only contract with Sussex, with his 151-ball innings containing 12 fours and a six.
George Tait (79) and Sravan Konidena (60) also played valuable innings against an attack again led by James Rudge who added three wickets to the six he took in the first innings.
Rain brought an early close in the Eastern Division One match at the South Northumberland club in Gosforth where Northumberland had gained the advantage over Staffordshire thanks to 93 from former Durham batter Harry Crawshaw.
Staffordshire led by 58 on first innings after Sam Atkinson (74 not out) shared a last wicket stand of 51 with Tom Brett (23) with Matt Oswell taking the second five wicket haul of his career.
But Crawshaw and his opening partner Sol Bell (40) cleared Northumberland’s arrears and, with excellent support from Alasdair Appleby (43) he then built a lead of 245 when the rain came.
There was another day of cagey cricket at Copdock where Cambridgeshire earned a first innings advantage of 25 which Suffolk had turned into a lead of 97 with seven wickets intact by the close.
Half-centuries from Callum Guest (66) and Alex Peirson (58) plus useful lower order runs from Mark Saunders (49) helped Cambridgeshire gain their first innings advantage despite a maiden five wicket haul from seamer Chris Steele and Tom Harper’s four for 63.
Suffolk openers Jack Beaumont (24) and Ollie Burle (27) began the second innings with a partnership of 61 but then fell in successive overs from Ben Claydon, a former team-mate.
Suffolk also lost debutant Ishaan Rakesh LBW to James Sykes before the close but Darren Ironside ended the day unbeaten on 32.
Cornwall tightened their grip on the Western Division title after they made Cheshire follow on at Redruth.
Rain wiped out the morning session and half of the afternoon but Cornwall made quick progress when play began by dismissing Cheshire for 184 with seamer Ryan Tonkin taking four for 41 and Ben Ellis and slow let-armer Elliot Hooper sharing the other six wickets.
Cheshire were 163 behind on first innings but reduced the deficit by 23 with all second innings wickets intact by the close.
Cornwall’s position was also strengthened by Dorset’s struggles against Shropshire at Whitchurch.
Former Glamorgan and Leicestershire pace bowler Roman Walker took seven for 38 – his third haul of seven or more wickets in successive matches – to dismiss Dorset for 98 in their first innings, with Ben Rogers the top-scoring with 17.
Shropshire opted not to enforce the follow-on but to build on their lead of 225 which they did with some difficulty and thanks largely to 69 from opener Harry Chandler who was ninth out.
A career-best five for 47 from seamer Matt Burton checked Shropshire’s progress but an inexperienced Dorset were left to chase 375 for victory.
Walker claimed Rogers as his eighth wicket of the match before Dorset closed on 70 for three.
A career-best 137 from Alfie Cooper kept Norfolk in the hunt for victory over Hertfordshire at Hertford in their final Eastern Division Two match of the season.
Cooper’s fourth Championship century of his career helped Norfolk to 317 all out a small lead of 20 with Ben Waring and debutant paceman Jake Whickham sharing eight wickets.
Hertfordshire ended the day 107 ahead with five wickets remaining after Andy Hanby took three wickets including Ben Clifford, LBW for 51.
Rain and bad light again meant short rations at Netherfield where a century from Matt Sempill helped Cumbria to 389 for six against Bedfordshire.
Sempill shared a fifth wicket partnership of 101 with Michael Slack before Freddie Fallows pummelled 52 not out from 18 balls, including seven sixes.
Bedfordshire were three without loss from three overs when the weather closed in.
